En este post les quiero contar sobre algunos de los medios de cultivo utilizados para el crecimiento de microorganismos, en este caso vamos a estudiar bacterias que a futuro se usaran en el análisis clínico en el laboratorio, con el fin de detectar enfermedades de naturaleza bacteriana y establecer diagnósticos confiables y seguros. La bacteria estudiada es la Enterobacter Cloacae Complex, un bacilo Gram negativo que puede encontrarse en el sistema digestivo 👀In this post I want to tell you about some of the culture media used for the growth of microorganisms, in this case we are going to study bacteria that in the future will be used in clinical analysis in the laboratory, in order to detect diseases of bacterial nature and establish reliable and safe diagnoses. The bacterium studied is the Enterobacter Cloacae Complex, a Gram-negative bacillus that can be found in the digestive system 👀.

Primero que nada, es importante saber que un medio de cultivo es una preparación artificial, cuyos componentes sirven de nutrientes para la proliferación de diversos microorganismos, pudiendo diferenciar unos de otros gracias a su comportamiento en los diferentes medios donde se siembran.🧫First of all, it is important to know that a culture medium is an artificial preparation, whose components serve as nutrients for the proliferation of various microorganisms, being able to differentiate one from another thanks to their behavior in the different media where they are sown🧫.

Agar nutritivo / Nutrient agar

El agar nutritivo es el medio más común compuesto principal por agar, que es un extracto de alga marina, en el que puede crecer cualquier especie bacteriana ya que tiene los nutrientes básicos para su reproducción, por ello primero realizamos la siembra en este medio para caracterizar las colonias formadas por nuestra bacteria en estudio.Nutrient agar is the most common medium composed mainly of agar, which is an extract from seaweed, in which any bacterial species can grow because it has the basic nutrients for reproduction, so we first performed the seeding in this medium to characterize the colonies formed by our bacteria under study.

Agar Sangre / Blood agar

El agar sangre es un medio enriquecido con sangre que tiene la finalidad de estudiar la capacidad hemolítica de nuestra bacteria. La hemólisis es la lisis o ruptura de glóbulos rojos del medio y esta puede evidenciarse por la formación de halos alrededor de las colonias bacterianas formadas, si hay hemólisis parcial se le llama alfa-hemolisis y los halos son de color verdoso, si hay hemólisis total los halos son translucidos y se denomina beta-hemólisis. La Enterobacter Cloacae Complex no presentó hemólisis por lo que se dice que tuvo una gamma-hemólisis.Blood agar is a medium enriched with blood that has the purpose of studying the hemolytic capacity of our bacteria. Hemolysis is the lysis or rupture of red blood cells in the medium and this can be evidenced by the formation of halos around the bacterial colonies formed, if there is partial hemolysis it is called alpha-hemolysis and the halos are greenish in color, if there is total hemolysis the halos are translucent and it is called beta-hemolysis. Enterobacter Cloacae Complex did not present hemolysis so it is said that it had a gamma-hemolysis.

Cled

Las bacterias tienen la capacidad de fermentar diferentes azúcares y producir ácidos, como glucosa, sacarosa, lactosa, etc. Para estudiar la capacidad fermentadora de las bacterias utilizamos el medio de cultivo Cled, que en su composición además de lactosa, tiene un indicador de pH llamado azul de bromotimol, que en medio ácido es amarillo y en medio básico es azul. Por lo tanto, las bacterias fermentadoras de lactosa van a producir ácido láctico y van a actuar en este medio evidenciándose por la formación de colonias de color amarillo, por su parte aquellas no fermentadoras crecerán en un medio de color azul. Nuestra bacteria es fermentadora de lactosa, sin embargo aun no había terminado de fermentar todo el medio al momento de estudiarla.Bacteria have the ability to ferment different sugars and produce acids, such as glucose, sucrose, lactose, etc. To study the fermenting capacity of the bacteria we use the Cled culture medium, which in its composition, in addition to lactose, has a pH indicator called bromothymol blue, which is yellow in an acid medium and blue in a basic medium. Therefore, lactose-fermenting bacteria will produce lactic acid and will act in this medium, evidenced by the formation of yellow colonies, while non-fermenting bacteria will grow in a blue medium. Our bacterium is a lactose fermenter, however it had not yet finished fermenting all the medium at the time of study.

Finalmente, se le realizó la tinción de Gram para conocer su morfología a nivel microscópico, siendo la Enterobacter Cloacae Complex un bacilo Gram negativo, fermentador de lactosa, con gamma-hemólisisFinally, Gram staining was performed to determine its morphology at the microscopic level, being the Enterobacter Cloacae Complex a Gram-negative, lactose-fermenting bacillus with gamma-hemolysis.

La importancia de conocer los diferentes medios de cultivos y su composición va a permitir predecir el crecimiento bacteriano y caracterizar a las especies, facilitando el diagnóstico de muchas enfermedades de origen bacteriano, por lo que el Bioanalista tiene un papel fundamental en el área de la Bacteriología 🔬❣️The importance of knowing the different culture media and their composition will allow to predict bacterial growth and characterize the species, facilitating the diagnosis of many diseases of bacterial origin, so the Bioanalyst has a fundamental role in the area of Bacteriology 🔬❣️
