with the capture device being the only item that is not guaranteed in each pack but a "must-have" to capture creatures once the game is live in beta
Will the backpacks be required as well? Or are they an extra holding space?

They won't be required, those are similarly like the mount meant to increase your bagspace as you're out traveling in the game and not at "home".
However this isn't 100% yet.
